Abstract

Blood agar merupakan media kultur mikrobiologi yang digunakan untuk mengamati pertumbuhan serta pola hemolisis bakteri, khususnya Staphylococcus aureus. Media ini umumnya menggunakan darah domba karena mampu menunjukkan pola hemolisis dengan jelas. Namun, keterbatasan ketersediaan dan tingginya biaya di Indonesia menjadi kendala dalam penggunaannya. Oleh karena itu, darah kambing Jawa berpotensi digunakan sebagai alternatif karena ketersediaannya yang melimpah di Indonesia, mudah diperoleh, serta lebih ekonomis. Penelitian ini menggunakan desain eksperimental dengan dua perlakuan dan tiga kali pengulangan. Tahapan meliputi defibrinasi darah, pembuatan media, inokulasi Staphylococcus aureus (ATCC 25923), serta pengamatan morfologi koloni, jumlah koloni, pola hemolisis, dan diameter zona hemolisis. Hasil menunjukkan morfologi koloni seragam (circular, tepi entire, elevasi convex) dan 100% isolat menghasilkan hemolisis β pada kedua media. Analisis statistik menggunakan uji Mann-Whitney menunjukkan nilai signifikansi sebesar 0,487 pada jumlah koloni dan 0,637 pada diameter zona hemolisis (p > 0,05), yang menunjukkan tidak terdapat perbedaan signifikan antara kedua media. Dengan demikian, darah kambing Jawa memiliki kemampuan sebanding dengan darah domba dalam mendukung pertumbuhan dan aktivitas hemolisis S. aureus, sehingga berpotensi sebagai alternatif media blood agar yang lebih ekonomis.
